Capsule OEM Formulation Design: Fill Composition, Glidant Choice and Fill Weight Matching

Capsules look simple: put powder in a shell. Anyone who has run a project knows the difficulty is not filling but filling accurately, filling consistently, and getting the plug to break up afterwards. Two formulations at the same 500 mg can behave very differently - one holds fill-weight variation inside 3% on the machine, another drifts past 8% and sits on the edge of the pharmacopoeial limit. The difference is usually decided in the first two design decisions. In capsule OEM, formulation has to be considered together with shell size and filling equipment; optimising them separately does not work.

Capsule OEM Formulation Design: Fill Composition, Glidant Choice and Fill Weight Matching

1. Back-calculate from the dose

Design starts with the daily dose and the count per day. If an active needs 600 mg daily and the plan is two capsules, each unit carries at least 300 mg; add filler, disintegrant and glidant and the total fill weight climbs well beyond that. The question is then whether the shell holds it. Standard two-piece gelatin shells run from 000# down to 5# by decreasing volume, with 0# around 0.68 ml, 1# around 0.50 ml and 00# around 0.95 ml. Bulk density for typical fills sits between 0.4 and 0.8 g/ml, so volume times bulk density gives an estimate of the achievable weight. If it does not fit, the options are a more concentrated extract, more capsules per day, or a larger shell - each with a cost, and all three belong on the table early.

2. Fill composition: active, filler, disintegrant

Filler adds weight and bulk while improving flow. Microcrystalline cellulose, lactose, starch and dicalcium phosphate are the usual candidates, chosen on hygroscopicity, compatibility with the active and cost. Extracts containing amine or aldehyde groups sit badly with lactose, where Maillard reactions gradually discolour the fill. Disintegrant makes the plug break apart once the shell dissolves: croscarmellose sodium, sodium starch glycolate or low-substituted hydroxypropyl cellulose at roughly 3-8%. Too little and the mass forms a lump in the stomach, delaying release; too much and moisture uptake rises, undermining fill-weight stability.

3. Flow is non-negotiable: glidants and lubricants

Automatic fillers dose by dosing disc or tamping pin, so flow properties set fill-weight variation directly. Angle of repose and compressibility index are the standard measures, with a repose angle below about 35° generally considered acceptable. Colloidal silicon dioxide at 0.5-2% is the workhorse glidant and magnesium stearate at 0.5-1% the usual lubricant. Two mistakes recur. First, magnesium stearate above roughly 1.5% forms a hydrophobic film on particle surfaces and actually slows disintegration and release. Second, adding glidant last instead of pre-blending it into the active means it clumps and distributes unevenly.

4. Matching shell size and filling method

Once the composition is fixed, the filling method is chosen. Tamping-pin machines rely on the powder's own cohesion to form a plug, and work best with moderately flowing, slightly cohesive powders. Dosing-disc and piston fillers suit free-flowing powders better. Granules and pellets are dosed differently again. The shell itself must suit the fill: hygroscopic blends do better in HPMC plant-based capsules, which carry less water and resist moisture migration; aldehyde-bearing materials are unsuitable in gelatin, because aldehydes cross-link the shell and delay dissolution.

5. Moisture and compatibility risks

Water migrates between fill and shell. A wet fill softens and tackifies the shell; a very dry or strongly hygroscopic fill pulls water out of the shell and makes it brittle. The practical target is a fill moisture of roughly 3-6%, with shell appearance and disintegration tracked in parallel during stability work. Fills containing organic acids, strongly acidic or alkaline salts, or high levels of surfactant can also destabilise the shell; these formulations need a one-month compatibility study at 40°C/RH75% during development rather than a surprise at commercial scale.

6. What sampling must verify

A sample is not approved because it looks right. A proper sampling run covers at least six things: appearance and shell integrity, mean fill weight and variation, fill moisture, disintegration time, flow parameters, and a short accelerated stability read. Fill-weight variation should be assessed on no fewer than 20 consecutive units, since mean weight alone hides the spread. With those six in hand the formulation can be judged ready for scale-up; without them, the risk simply moves downstream.
