Capsule OEM Filling Control: Machine Setup, Dose Variation and In-Process Checks

Capsules are the format most sensitive to fill-weight accuracy. Two batches both labelled 500 mg per capsule can differ by 8 percent, and at the mild end that is a labelling risk, at the worst it means under-dosed product and complaints. Many brands blame the machine, but the root cause is usually powder behaviour and the order of setup. This guide explains how fill weight is actually controlled on capsule OEM lines, and where it most often slips.

Capsule OEM Filling Control: Machine Setup, Dose Variation and In-Process Checks

1. Capsule fillers are tuned in a fixed sequence

An automatic capsule filler runs a chain: separate the two shell halves, orient cap and body, dose powder into the body, reject rejects, lock, eject. Setup must follow that chain. Step one is separation rate - get empty-capsule opening above 99.5 percent. Step two is the dosing disc and powder-scraper gap, which sets the base weight. Step three is the full run. The common mistake is loading powder, running the batch, then tuning weight afterwards; reject rates climb and the dosing disc never fills properly, which amplifies variation.

2. Dose variation has a legal limit, then an in-house one

Hard-gelatine capsules are judged by a weight-variation limit: plus or minus 10 percent below 0.30 g average fill, plus or minus 7.5 percent at or above. Plants normally hold tighter, controlling individual-capsule spread to plus or minus 5 percent of the mean and sampling at least 20 capsules per batch. Note the judgement uses the measured mean, not the label claim. A sample mean inside the limit passes, but if the single-capsule range is wide, a customer audit will still call it unstable.

3. Powder flow is the hidden variable

The same machine can drift when a finer powder batch arrives. Poor-flowing material (high angle of repose, bridging) under-fills the dosing pockets and feeds erratically, so individual weights swing. The usual responses are a glidant such as colloidal silica, holding humidity at 45 to 55 percent RH, and dry granulation where flow is bad. When the active load is high and flow is poor, flow should be a first-article acceptance item, not something discovered in production.

4. First-article and in-process checks must be linked

The sound release path is three gates. First-article: take the first 20 capsules, weigh them, only start the batch when all sit inside limits. In-process: pull 10 every 15 to 30 minutes, stop and re-tune on any drift. End-of-batch: re-check the tail to confirm stability across the whole run. Omitting the in-process check and finding the problem at finished-goods inspection is where most disputes start, because by then the batch cannot be corrected.

5. Four practical points on linked control

One, match dosing-disc speed to scraper pressure - faster rotation under-fills. Two, misalignment between the lower module and dosing disc beyond 0.2 mm causes periodic weight cycling and needs recalibration. Three, seasonal humidity changes the bulk density, so re-run first-article at season change. Four, for poor-flow formulas prefer a machine with a more stable locking action rather than chasing throughput at the cost of weight stability. Bind these four to the three gates and variation stays inside plus or minus 5 percent.
